Lecture 3

I have to digest tissue at 37 deg. at 1 hr - but want to have more time, so I do it at 18 deg. and draw it our longer - do it for one hour

Place in sterile plate TWO days, with culture medium present.

Must shake it while heating in incubator. Trypsin Works like butter onto an English muffin - gets trypsin into "nooks and crannies" between cells, shaking helps is get in. As a little "gets in there" - it erodes away the most exposed proteins that link cells together. Might not get 100%, but gets a lot of the linkages.

PRIMARY EXPLANT is all up to step 3. Because we're "explanting" tissue.
